Abstract

The breast cancer cell line project was setup to provide and share for the fellowship of Institut Curie, cell lines from ATCC annotated with their genomic, transcriptomic properties and their sub culturing characteristics. In this aim, 35 cell lines were established from new vials from ATTC, aliquoted and frozen. The cell line collection was composed of normal cell lines (MCF-10-A), modified cell lines (htert HME1), and tumor cell lines. Frozen vials are available for research for a low cost. Details of sub culturing, transfection are available to facilitate and standardize the use of those cell lines. Genome analyses were conducted using Affymetrix SNP arrays to define copy numbers, allelic imbalance, LOH, and to identify genome alterations per cell lines. Other analysis such as hot spot mutations, will be implemented to share all results that belong to cell lines Transcriptome analyses were conducted using Affymetrix Exon arrays allowing quantification of transcript by an average of 40 probes. The data analysis was performed by Genosplice technology and is available on the Easana website (www.easana.com) to visualize gene expression per transcript, and per exon to highlight splicing events and more. The portail will propose further analysis when they will be available (miRNA,..) This project was supported by Institut Curie (PIC Cellular Models, Clinal Scenario and screening), the Translational Research Department (CEST), the genomic platform, and Affymetrix, based on F. Radvanyi request in 2009. In the translational research department, cell lines were amplified by E Henry, V. Maire, B. Marty Provost in the Breast Cancer basal like Group, headed by T Dubois. Genome and transcriptome analysis were performed by E Henry, C Reyes, C Hego in the genomic platform managed by D Gentien.
